Located in southeast part of Ulster County, the town of Marlborough was named after the Duke of Marlborough. U.S. Route 9W passes through the eastern part of the town while US Route 44 and NY 55 cross the northwest corner of the town.

Marlborough is known for its stunning river, valley and mountain views.  It is made up of several communities and hamlets: Baileys Gap, which is a hamlet at the north town line on Route 44; Central, a location west of Milton; the hamlet of Marlboro, which is in the southeast part of the town on Route 9W; Milton, a hamlet in the northeast part of the town and adjacent to the Hudson River; and Lattingtown, a hamlet in the western part of the town on Route 11.

Among the historic sites in the town are the Lattingtown Baptist Church, which is listed on the National Register of Historic Places, and the Gomez Mill House, an historical site and the oldest Jewish residence in Ulster County. It was the first paper mill in Ulster County.
